I have created a powerpoint with "On Click" animation (which I rarely use, I usually use "with or after previous" timings.)
When I preview the slides in presenter, I can't click to bring up the next animation. The annimations just play automatically as if I had used "after previous". By the way, originally the timings were "after previous" by my reviewers are requesting "on click".

If issues persist, you may also want to take a look at running through some of the troubleshooting steps listed here and let us know if you find improvement or you continue to have difficulty.
I looked a little deeper and it looks like presenter just doesn't support the animation on click. I do not have any audio I am synching to. I just wanted to allow the user to control the pace of the information being presented on the screen as opposed to setting a specific amount of time between information appearing on the screen.
My workaround is splitting the slides into multiple slides. - this would be a really nice enhancement to support the animation on click functionality in powerpoint.
