Is there a way to create either a Master slide or template for consistent formatting of question slides?
I have reformatted the "question slide" in slide master, selected insert essay question, for example, and then changed the layout to the "question slide" from slide master, but the formatting changes from the "question slide" master slide do not take place.
I have several questions that will require the same formatting, in multiple instances. Is there a way I can create a master slide or master style for question slides?

As you noticed, you'll want to customize your slide master layouts in Storyline, so that the layout you use for quiz questions contains any content that you want to appear on all quiz slides. You can learn more about slide master layouts in Storyline in this tutorial.
Can you tell me what content doesn't show up on the quiz slides? Is it normal formatting or possibly formatting that would be set through the Feedback Master?
