I can see the behavior you're describing in slide 1 where the shape is a pentagon, but I'm not sure how you applied the shape, as I'd like to see if I can recreate the behavior in a new file. Could you share a bit more information about that?
I tried this in my own files with a jpg and a png image, and all was working as expected in my Storyline file. I've attached it for you to take a look at and please let me know if this one is behaving as expected for you.

After I inserted the image and selected it, I went to the 'Format' tab and then selected a shape from the drop-down menu, 'Picture Shape'.

If so, you'll want to confirm that you're working with all local project files, as working on a network or USB drive could cause unusual behavior, file corruption, etc.
If the behavior continues for you while working on a local drive, please also follow these steps to conduct a repair of Storyline, and also check that you're using update 5 of Storyline.

The file you uploaded is behaving as expected.
I was only changing the transparency of the picture, not recolouring it- should it work if you are only changing the transparency?
