I've looked at your file and I don't have any trouble clicking the buttons more than one to get the layers to display. Maybe I misunderstand what the issue is.
I am looking at this slide in preview mode and I am able to click on the layer buttons over and over and the layers show every time. Am I doing the wrong thing? Is that the issue you are having?
It looks like you are leaving your layers open and this is preventing user from clicking same button twice in a row. I added a close layer button on each layer when the timeline ends and I think it's working as you intend now.

Hi Sharon, are you able to share your source file at all? It would help to see it.
When you say "once show it doesn't come on again" do you mean the button or the layer?

Yes this is fine, thanks , but if you try and repeat clicking on the same button twice:
eg: Physical reasons > physical reasons
