I have a slide with 16 layers on it with videos (each layer has a video - appx. 15MB to 60MB). Does this affect the performance of loading the slide and playing the video?
I'm not seeing anything attached here. Did you check out the information that Matthew shared above on loading? That does seem very media heavy and some delay during loading does not seem unlikely.
16 videos on slide 3 is not a great place to have them as Storyline will preload 3 slides before starting. You may need to split these into separate slides
So does it makes since if add a new slide 3 with some instructions and have on slide 4, videos, will it help? Or you recommend to break the layers in to 16 slides?
It won't help a lot as you have minimum of 240Mb of videos (likely much more) that will need to be downloaded, if you plan for this to work on a tablet I would split anyway,

At the 3rd slide I have 8 layers with 16 videos all together, so does this causing the slowness?
So instead of layer, I need to have slides?

16 videos on one slide sounds like a lot to me.
