I got stuck with audio play/pause button and with timeline. I could be able to play/pause audio along with the time timeline, but my requirement is to control only audio let the timeline run in the background and when I resume(pause to play) the audio button it should start from where the timeline is running.

If you mean at 10 seconds, you pause the audio, and when the timeline reaches 20 seconds, you resume the audio, and it plays from where it was (10 seconds into the audio), then yes, this is easy.
If you mean the audio should resume and start playing from where the timeline is (20 seconds into the audio), then no, you can't do that.
I would like to know one more thing, that I have created a custom play/pause button where I have added a trigger to the audio. And the audio is not playing inside the slide but when I preview its working fine.

If you have a dded a trigger to play and apse the audio, you likely need to add a trigger to play the audio on timeline start. If the waveform is no longer visible on the slide this indicates the audio is no longer timeline dependent.
Here I am attaching the .story file. In chapter 1 slide 1 I have added a trigger to .mp3 (audio file.) and I am not able to hear the audio in timeline after adding the trigger to sync the basic animation.
Here's what happens when I preview your story. Whether I preview this slide, or the entire project, I continue to hear the audio clearly.
As you can see, when the slide starts, the audio plays. When I click the mute icon, the audio stops. When I click the mute icon again, the audio continues. I think this is the action you describe that you want. Are you seeing something different?
I do notice that when the audio stops, that the timeline (and thus the animations) continues. If you want the slide timeline to pause, then you need to duplicate these triggers on Chapter 1 Slide 1:
You will need to change the new ones to pause and resume the timeline on the slide.

Lots of bad experiences have taught me that you can use playing from the timeline to synch the animations only for very crude approximations. If you need any degree of accuracy with the animations, you need to play the audio through Audacity or MovieMaker and record the moments when you need the animations to occur, then transfer those to SL.
If the animations and audio are synched, he still needs triggers to pause the timeline when he pauses the audio.

Not sure what you mean by this, but all that really matters is what it does when you publish it to and play it from its intended target.

Adding a trigger to play the audio will take it off the timeline, the audio will only play on the timeline when it is shown as a waveform
