How do I set up my mouse and auto populated rectangle that displays with each click to be the same every time I capture screen shots? I am tried setting the mouse on the first slide to be the default and the shape to be the default but that didn't work. Every time I capture slides it puts in the default mouse with the red flashing circle and the blue rectangle.
I'm unsure that I'm following what is happening with your file. Are you using the screen recording behavior of Storyline? You can't modify the default captions as you'll see here - but there are a few ways to change them after.

Currently, Storyline puts in the default hand curser and blue highlight box every time I capture. I have tried using "Set as default" but that doesn't work. It is a pain to have to go through each and every slide to change the mouse and highlight box. Any help would be appreciated.
Have you looked at modifying your theme colors to adjust the caption/highlight box? You'll see a similar discussion here on what you may want to look into changing, and a tutorial on theme colors here. I don't believe there is a way to set a default mouse cursor, but you could use the format painter to make things a bit faster.
