I have search around and not seeing any major answer to this. The little preview play button in the timeline. I am trying to listen to my audio so I can add cue points. When I select it, the mouse icon just spin around then nothing happens. This is SL2.
The play bar should still work to play the slide, so if it's not you may want to look into conducting the repair of Storyline to resolve any odd behavior. Additionally please also confirm that you're working on local project files as described here.
I did re-install the program. The play button will work, but it slow. It act as if it buffering before the side play. I am working off of my laptop hard-drive. Not using usb, external drive, no long naming, etc.
Have you checked to see if any other programs or applications are running in the background that could also be interferring with the overall memory available for Storyline? You'll want to make sure that the computer you're working on meets at least the minimum system requirements here.
Also, is it just that one slide with audio that is slow, or is it all your slides?

Just found out on my own...if I hit the spacebar, like in audio editing, it play the audio from the timeline.
