I'm trying to create a way to make it clear to users that they must click on the video to make it open in a new window.
For that purpose I'm trying to put a fake playbutton on top of the video. BUT, is there any way to make the button visible but still 'transparent' so that the user is in fact clicking on the video?
That's what I thought, but it doesn't work because the video is supposed to open in a new window. For some reason a trigger does not open the video in a new window.

In answer to your original question, I don't think that a shape can be semi-transparent AND invisible. Unfortunately I don't have a usable solution for you - sorry - but I'm wondering if adding a trigger to change the state of the play button to hidden when the user hovers the mouse over it (and have it restore on mouse leave) would work? In theory, the play button is visible, so the user can see they need to click on it but by hiding it on mouse over, they actually click on the video.
What is it that triggers the video to open in a new window? Could you make an image that was the thumbnail from the video you wanted with a play button on top and then have that entire thing be the trigger to open it in a new window? Alternately if the place the video is hosted (if external) lets you select your thumbnail could it be a separate image (again with a play button superimposed on it)?

Can you not just add a trigger to the play button to play media when the user clicks on it?

I think the mouse over might confuse most of the users, but it may be the only solution.
Unless there are other ideas out there?
