I'm sure I must have a box checked where I shouldn't...but why would the volume mute during a screen recording (with audio) when the user clicks anywhere on the screen?
Currently, the forum only supports files that do not exceed 20 MB. If your file is larger than this, I recommend importing the trouble slide or slides into a new project. You can share that smaller file here with us.
It looks like the problem may be that the user is interacting with the video. It doesn't look like the volume is actually muting when the video is clicked. Rather, the video is being paused.
For example, if I click on the video that's in your slide in the file you shared, it does pause and the audio stops. If I click on it again, it continues where it left off.
In order to get around this, I recommend placing a large object on your slide and changing that object's transparency to 100%.
I've done so in the project you shared, placing the transparent object over the video, but behind the interactive elements on your slide (the rectangles and hotspot). If you take a look, you'll see that clicking on the slide does not impact the video and the learner should still be able to use the other elements in the slide.

Quick question, how did you change the transparency? I've put in a rectangle over the full slide and then set the fill to none, but it didn't work.

Right-click on the object and select "Format Shape". From here, you'll want to adjust the Transparency for the fill of the object:
