If you publish to web then click the story.html5 file in the output you should be able to preview the html5 version in a supporting browser such as chrome or firefox.
There is no way currently of simply 'previewing' the html5 content without publishing.
As Harri suggests, you have to publish the output in order to preview the html5 - you can review the html5 on supported desktop browsers without uploading to a webserver by opening up the published content folder and dragging the story.html5 file to the Chrome, Firefox or Safari browser on your desktop, but you have to be a little careful with this because the implementation of html5 on the mobile Safari is different. So you might find html5 works fine on your desktop browsers but still have issues on mobile safari.
You might be able to install a mobile safari emulator on your computer. Here is a link to some info on that. I have not tried it so am not sure how well it works.

If you don't have an emulator then the only way to test is by uploading to a server and then viewing on the iPad.
