I would reckon, on many LMSs that if people can actually navigate through to the point where they get to, and run the course, then they have probably passed the literacy test :)

The first thing is to get your questions sorted. I reckon that this topic would have HUGE potential for all kinds of interesting options, "Click on the area where..." (using hotspots), "Select the order in which you sign on....", (or whatever).
I think Studio or Storyline have the tools you need as standard, but the hard part is the test questions themselves, and tying it all to a "real-world" reason for the learners.
