You could create a T/F variable that's set by the hotspot on slide 1.1 that defaults to F. Set it to change to T when the hotspot is clicked. Then on slide 2 set a trigger to show layer 2 when the timeline starts if the varialbe you set on slide 1 is true.
Forgot to mention that on slide 1 you need to make sure the variable happens before the trigger to go to slide 2. So you interaction should go like this:
Create a T/F variable and set it to F
Add trigger to your hotspot that changes the variable to T.
Add another trigger to your hotspot that advances you to slide 2. (This must come after the variable trigger change.)
On slide 2 set a trigger to show layer 2 when the timeline starts if your variable if T.

No you cannot move the Hotspot you have to add a new hotspot for every slide.
Also you can try using triggers instead of hotspot. if you need help you can ask me anytime
