I am not sure to be honest. Here is a link to the 508 standards guide. Not everything in Storyline is going to be section 508 compliant. Just primarily those things listed in that link from my earlier post.
2 year old post, but I'll necro it with my 2 cents lol, hotspots ARE NOT 508 compliant in storyline 2, they were working for me in storyline 1 (which you were probably using at the time of the question) but for future threadviewers I'd like to mention that instead of a hotspot object, for 508, one would want to use a rectangle with no fill or outline, and if that 'hotspot' does not have any triggers on it you will want to give it a 100% transparent fill, or storyline will not add it to the tab order.

Not really. You can use them but you need to provide an alternate interaction as well. Here is the link about Storyline and 508.
