Unfortunately, the answers to your questions are by default not editable. I could be wrong but the only editable region I can find is the Review layer's master which only pertains to the rounded rectangle and text area. Everything is driven by the Quiz mechanics.
However, with Storyline anything is possible!
It takes a bit more work and design plus your comfort level with variables. Essentially you can recreate an entire Quiz, Feedback Layers, and Review from scratch and customize the look and feel anyway you want. By taking this approach you will invest a considerable amount of time but the flexibility allows you to do even more than what the default quiz does.
I mean the quiz review function not the results slide - where it shows what answer you selected and adds those nasty correct/incorrect layers at the bottom of the screen. Can't see where those layers come from - are they editable?
I know this is old, but you can remove the box by right clicking on the slide in the left panel and changing the layout to a blank slide. Make sure you're on the layer that you want to customize when you do this.

When you say "custom review" are you referring to quiz feedback or the Results slide?

You can customize the review feedback slide. Here is some information on how to achieve this. It works the same with Storyline 2.

You covered the correct/incorrect coloured boxes with another box. Is there a way to turn the boxes off?

Unfortunately there is not a way to turn that off. you would need to cover it with another object.
