I have a 20-slide course for which I recorded the entire audio in one take, as 1 large audio file. Can I import the audio file into Storyline 2 and parse it out slide by slide, or would I have to edit the audio file into segments first, per slide, and then import into Storyline?
At some point you'll have to edit the sound file. You can edit it in some other program, or copy it to each slide within SL, and then edit each slide's portion in the audio editor (double click the sound on timeline) Good Luck!

either way you'll need to edit. Storyline can do it using the options > audio editor.
I would start from the last slide and work forward.
