I hadn't seen that error message before, so I asked a few of my colleagues and it looks like it's a Windows error ("Access denied because another caller has the file open and locked.") I'm glad that the "save as" worked for your project file.
This happened again this morning. All I had open was Storyline (my project file), Word (storyboard file for course), Firefox (one of the Articulate forums), and Audacity (an AUP file with some of the narration used in the course).
I had previously (couple of days ago) exported a WAV file from Audacity, and I had just imported it and another WAV file (from the SME) into one of my project's slides.
On a whim, I just closed Audacity and then tried to save my Storyline project again. Save worked just fine. Weird . . .
I haven't heard of Audacity causing issues with Storyline when both are open, but stranger things have happened I guess. I'm glad you were able to save in the end!

Thanks, Ashley.
Interesting. Not sure what other file was locked. Oh well, good to know it's not Storyline.
Glad I could help point you in the right direction.

Interesting, Gerry. I occasionally have Audacity and SL opened at the same time, but haven't yet seen that one.
