You'll want to save a copy of the .story file, and I'd suggest doing so before publishing just to be safe. If you publish a course what you'll see is a new publish window generated where you can zip the files, open the publish output folder, etc. You'll still be within the Storyline program, and you could still save the .story file at that point. If you have a published course but do not have the original Storyline file, there is no way to revert the published file into a .story file.
