As long as you retain the original story file you can still make any edits you like. You will have to republish the course though. You can not make changes directly to the published course.
What do you mean by not making changes to the published course? Can I not pull up my published version, make my changes and then publish it with a different name?
A published course is either a HTML(Web) course or a Scorm package. Scorm is a zip file you cannot edit only upload to a LMS ( Learning Management System ). A HTML course you can edit as any HTML. But not in Storyline when its published. You need a HTML-editor for that...but its not really funny editing in the published output.
Storyline is the authoring tool... when done you publish it to either HTML or Scorm. When you want to change something... change it in your original files in Storyline...and publish again.

If you want to make edits to a project, you will need the original .story file, which is the file you save from Storyline.
You can open the .story file, change it, and then re-publish it for the format you need.
