It sounds like you inserted an image of dark-blue quotation marks. Like photos and other illustrations, those can't be recolored in Rise.
Here are a couple options:
Option 1: Insert the quotation marks as text. Adjust the size, font, and color as desired.
Option 2. Insert the same graphic in Storyline. Right-click the graphic, and Export/Save it on your computer. Change the color with image-editing software. Then insert that image.
For example, this file started as a dark-blue quotation mark. I used the Fill option in SnagIt to change it to purple.

You can save any block or set of blocks as a Block Template. That template can then be inserted in any Rise course. Here's more info: Rise 360: Creating, Sharing, and Reusing Block Templates
