It's a shame though about the slide master layers vs main slide layers.

Thanks for reaching out and sharing your file with us! I looked at your file and noticed the same behavior you mentioned.
The layers included on a slide will take priority over the layers that are on a slide master. That is why you cannot see the master slide layers when previewing your course.
