Is there a program/app that will review the readability/assess the tone of your project's font, spacing, and structure? Like a Grammarly for font/spacing/structure?

What you're looking for is a typography calculator or font size calculator. There are dozens (probably more) of these tools. A couple I've used in the past include:
Most of these tools are designed for the web, so your measurements will be in percentages or ems, but you can use the output to estimate in your authoring tools.
