Take a look at the article below for tips on how to use a screen reader with your course:

What I suggest doing first is checking the focus order of the slide that has the buttons and layers to make sure it's following the correct order and that the text in the layer is also listed:

    I'll take a look. I think it's more of a matter of figuring out NVDA.
