I have used Callouts to indicate what a learner has to do in a Try me. The callouts seem to appear correctly when I "Preview" a slide. However, when I publish the file, the callouts do not appear.
There are no Transitions or Animations for these.
Is there a setting for this to appear?

When you published your file, which format did you choose (web, LMS, Review 360, etc.)?
If you're comfortable sharing it either publicly here or privately through a support case, we'd be happy to take a look at your .story file to see what's going on.
I was not viewing the Published page correctly. There is a left Navigation pane with different scenes, and I was always viewing the topmost scene, which is the original recording. Obviously, the original recording does not have any callouts.
