I am trying to create a lesson where learners will be asked to identify correctly several different elements within a graph. I would like to use audio to prompt the learner (i.e. Identify the axis that shows hours per day) with the question then upon selection there will be a response (Yes, that is the axis. You can see here... No, that is incorrect because...). If I am using the same graph can I make this one slide with multiple layers or should I use multiple slides?

I would definitely go down the layer route.
I would also break up your audio in to different clips and use triggers to play the appropriate audio track based upon the feedback you want to give.
You could alternatively use states....but I personally prefer using layers for this kind of thing.
