trouble with markers and variables

Another noob question from me. I'm trying to make a navigational slide using markers, layers, and variables. The idea is that the learner will click the pulsing marker and that will reveal a brief lecture. Once the lecture ends, the first marker disappears and a new pulsing marker appears on the graph, which upon clicking will reveal the next lecture (and so on). I tried to accomplish this using variables but failed. I also tried to use a trigger to change the state of the marker to "disabled" by making the marker transparent but the pulsing continued. Any help would be greatly appreciated.

I wish there was an option have the markers automatically play through. My users don't want to click through and just want to watch. Or give them an option to just play through all the markers. So opening and closing each marker as it's done with the audio.

Markers by default have to be clicked or manually activated - but you could set up some custom shapes that look like markers but are set to open(change their state) based on a timeline trigger.
