The color of your markers on a Labelled graphic is controlled by the Theme color you set within the course settings. All the markers will draw from that one color, but it's a neat idea to have multiple colors. I think it would be worth telling our product team about it here!
The spacing of the markers is entirely up to you! You can click anywhere on a labelled graphic to add in a new marker, and then click to drag it to a new location if you need.

I too would like to be able to use multiple accent colours. In our branding, we have a primary colour and a secondary colour that we use in various instances.
