Try using the Project.ElapsedTime variable listed in this article in your course, and let me know if this is something that you can use for your calculations.
An easy way to get an estimate is to click the Publish button, then select LMS/LRS tab, then click the ellipsis button to the right of the Title field. The estimated duration shows in the Duration fields.
There is no way to know for sure. All the calculation methods rely on the timelines of the slides. A menu slide, for example can have a timeline of 5 seconds, yet be visited 10 times. A slide may have content on layers that stops the timeline.
The only really accurate method is to time it, perhaps during the final Quality Control pass. That is, if you do QC.
