Is there anyway of setting a default state (throughout an entire course) to the built in Next/Prev buttons in 360? I want the next button to be 'disabled' at the start of each slide, and then change to 'normal' with the use of a trigger.
More for timesaving reasons, I wonder if there is an easier way to do this rather than create a trigger on each slide to set to 'disabled' when timeline starts?

You can set the navigation restriction for your entire course using the built-in options within the Player. You can read more about that here.
I recorded a quick Peek 360 video to display that option along with the copy/paste feature of the triggers if you decide you still need to create custom ones per slide.
