I want to have the audio that plays when you open the scene to stop when you click on the new layer. Currently the default audio and the new layer audio play on top of each other.
I tested your slide and it is possible to cancel out the base layer audio once you click the hot spot 'click for sound' ^^ Michael had it right. On the layer you wish to have override the base layer, goto the properties of that layer and click pause timeline of base layer. That tells the base layer to stop or pause and the new layers audio to begin without them overlapping....Good call Michael.

Try setting the layer's properties to 'Pause timeline of baselayer'.
