Yes we can lock the drop target. Attached example will give you an idea about locking dragables. Can you please have a look and don't hesitate to leave a message if you required any further explanations.
You can see the built in drag and drop options that you can enable here and if you were to add in your own additional triggers to do thing such as track a variable (how you could tally the number of drops) or similar it may override the built in drag and drop items, so you'll want to be cautious of that.

I forgot to ask if it's possible to lock a drop target after 1 item got dragged into it.
