Let's go over your questions and possible solutions:
1. Question 5 is a drag and drop and unless I select the correct answer I have to click on the submit button twice.
This is related to the number of attempts allowed:
If you have more than one attempt allowed, when the user chooses the incorrect one and submits, they have a chance to try again, but no message is given since you don't have a Try Again layer.
I fixed it by changing the number of attempts to one.
2. When I go to retake the course I can't click on any of the answers.
I couldn't replicate this issue, as the retake course allowed me to click on new answers for each question. However, this could be related to the issue below.
3. The success layer is not showing when I pass the quiz.
I believe there was some corruption with your Results slide. I was only able to get it to work once I deleted it and re-created the slide.
I am attaching the edited .story file with the changes above.

Oh that is weird, I wasn't getting an errors indicating that is was corrupt but I'm glad you caught that.
