Put a variable at the end of layer 1 to indicate that layer 2 may now be visited.
On the base trigger that shows layer 2, put a condition that it be shown only if the slider is the corresponding number AND the variable is set to true. This gives you the added advantage of allowing users to go back to any layer they have already visited.
I am not aware of a way to lock the slider itself, but using the method Walt suggested, you can stop the layers from showing until the previous layer plays. See attached - I added a T/F variable at the end of each layer. (I only did base, layer 1 and layer 2)
You can see that the slider can still move, but layer 1,2,3 will not show/play until the prior ones are complete. Since the slider can still be moved, I am not sure if this is the best design approach for locking layers. Hopefully others will jump in with some suggestions.

Walt, I'm not following you. Wouldn't it be a trigger not a variable?

Good point Emily. I'm thinking of taking it out. It seems like it would only confuse the learner.
