I have an audio introduction on my main menu layer and want it to play only the first time a user visits the page, not each time they return from a sub layer. Is this possible?
This file shows how you could set up what you're asking for. There might be a more elegant way, but I have it so that if you go to another lay, or another slide and return the sound does not replay, only on the original entry. You could run into a problem if the person does need to replay the sound, but then again you could add a replay audio button if needed to resolve.
Do we have the feature to activate the same trigger in presenter ? I have the same problem that if the user returns to the page its playing the audio again. But i want it to play only once. Please help.
That's not an option currently available in Presenter '09. You can find a couple workarounds here, but it's not something that can be set out of the box.

Thank you so much Brian! That works equally well if you have a sound you don't want to play at all except when triggered.
