I am not sure what I am doing wrong. I want slide 2.2 to work like slide 2.1. Where you switch to the layer, the there is only 1 audio on that layer and the box changes to visited. Please help.
You had copies of the three buttons on all three layers. By the way the triggers to change button xyz to Visited are not necessary. The Visited state will automatically be displayed when a button is clicked. Have a look at the attached.
I see that the text from Slide 2 is missing although it's viewable on the slide itself - and I'm not seeing anything to hide it, but Michael may have done something that I'm just not seeing yet? You also have copies of the text on the layers too - which I'm unsure based on that if you want them to show on the base slide and on the layer itself or just on the layer?
As Michael said, you have copied the buttons from the base onto each slide. The buttons on the base are changing to the visited state (turning red) when clicked, just like you would expect, but you cannot see that because the copied buttons in the layer are on top of the changed buttons. Delete the buttons in the layer and then you will see that the buttons on the base are working properly. The first graphic below shows this for you.
You cannot see your text because you have the Initial State of the text boxes set to Hidden. You never change the state of the text boxes to Normal, which would make them visible. To fix this, change the Initial State of the text boxes to Normal (not hidden). The second graphic below shows this for you.
So a quick summary of what you might consider doing...
Delete the buttons on each layer. You only need them on the base.
On the base, delete the triggers that say "Change button X to visited..." . You don't need them, as their state will change to Visited automatically when the learner clicks on them.
Delete the text boxes from the Base, as I assume you only want them to appear when the appropriate layer is shown.
Use only the appropriate text box on the appropriate layer. For example, on the TA layer, you only want the text box that pertains to the TA discussion. Delete the other text boxes off of that layer. Do the same for the other layers.
Change the Initial State of each text box to Normal - they will only appear when the layer is shown, so they don't need to be Hidden.

Another option is to keep the learner from clicking and launching the layers until they have heard all of the audio on the base, which may be what you really want after all? You want to be sure they have heard everything before they start clicking around to the different layers. If so, then in the timeline on the base, don't show the buttons until after the audio completes.
An alternative method is to show the buttons right away like you do now, but use a hotspot with no trigger over the top of the buttons so the learner cannot click the buttons. Then remove the hotspot once the audio has completed by having the hotspot "end" at the end of the audio in the timeline. But the first method is simpler. :-)
