I've added TTS clip to a slide that I'm working on for a course. However, when I hit the play button on the lower left side under the timeline so that I can add cue points, it will not play. It's very strange. If I click to preview the scene or slide, the audio plays. It just won't play in the slide.
I'm not seeing that behaviour. When I click the play button I can hear the TTS and add cue points. I tested it a couple of times. Where do you have the .story file - are you working on your local drive or from a network drive?

My experience was the same as Wendy's: the audio file worked when previewing it on the timeline, and I could add cue points.
The first step I'd recommend is repairing Storyline 360:
