I often use the Spotlight & Magnify effect in Snagit after I take a screen capture to emphasize sections. For example, if I had three items I want to emphasize at different points, I'd make three images spotlighting and magnifying each section in turn. Then I'd layer them on my Storyline 360 slide and have each one display at the appropriate time on the timeline.

From what I understand, maybe you could try using shapes to cover the parts you don't want to highlight.
You can use black shapes with some opacity so that you can still see through but it should emphasize the parts that aren't covered.
And with the timeline, you can show/hide parts of the pictures at different timings.
