It is definitely possible. Storyline has triggers that allow you to "Pause media..." and to "Play media..." based on certain activities or parameters.
What I usually do is to place a 100% transparent object on top of the video (same size as the video) that has a trigger on it to "Pause timeline" when the user clicks on the object. You can then either do a state change of this object, or call a different layer (maybe called Paused) that will "Resume timeline..." again when the user clicks that object again.
