I don't understand but I have made some changes to previously saved file in the audio but the published course doesn't play the voice over of the new added voice over. Thought it plays after I click on the reset button.

Did the changes that you make include edits to triggers/variables?
Using the same audio configuration, are you able to replicate the behavior in a new Storyline 360 project file?
Additionally, would you be willing to share a copy of your project file here or in private by opening a support case so we can take a closer look at what's happening? We'll delete it when we're done testing!
