I have created a sequence drag and drop activity and am having some issues the numbering disappered when preview the slide. and also how to customise the layout in term of colour and shape size
I was able to reproduce what you described above when testing your project, and also in a new file, so I logged it as a possible software bug to our team and linked this discussion to the report.
I want to share some information on how we tackle bugs, and we'll let you know as soon as we have an update on this issue.
There isn't any way to customize the appearance of the numbering in sequence drag-and-drop questions, but this sounds like an awesome feature to have. I’ll be happy to pass your thoughts on to our product team, but you can also feel free to detail them more through a feature request!

I have some great news to share. We just released another update for Storyline 360. In Update 67, we’ve included important fixes and new features.
One of the fixes we’ve included is:
The numbers were missing from sequence and ranking drag-and-drop questions.
To take advantage of this update, launch the Articulate 360 desktop app on your computer and click the Update button next to Storyline 360. You'll find our step-by-step instructions here.
