I am creating several "Quick Choice" interactions. I am adding audio to the "home" slide of each interaction, plus audio for each choice. The latter play fine but the initial audio (with the instructions to click the answer) doesn't automatically play. Then it seemingly plays at odd times, after answering one or other of the choices. Why would this be? How can I fix it?

It sounds like you are running into an issue with the auto-play of your course, prior to any interaction. This is the expected behavior from browsers now.
Will these interactions be a part of a bigger project that may have interaction prior to arriving on this slide? I'd be sure to test in that environment.
